I'm getting ready to poke about in that a bit, but I think I have an idea of one of the problems.
I know a list using markup was a problem in GTK 2.x... might be for 3 also.
The list was already using markup (to get the font sample). One thing that did appear to slow down start-up was adding the separator function.
One aspect I missed with my earlier tests: AFAICT it's not just affecting launch time, it also noticeably delays opening additional document windows from within the running inkscape instance (starting with r12105).
