If you mean like the "XXL" rectangle in:

then here is one way:

1. Create the rectangle and set its color
2. Create the text and move it into place over the rectangle (use the alignment tool to make it perfectly centered)
3. Select both the text box and the rectangle
4. Use the menu item Path >> Difference (or press Ctrl + -)
